Bleomycin Separation

This illustrates the chromatographic separation of Bleomycin sulphate, a mixture of cytotoxic glycopeptide antibiotics used in the treatment of various types of carcinoma.

The USP assay procedure defines a 60 minute binary gradient followed by an isocratic elution.
The separation can be greatly accelerated using a much steeper gradient (30 min) followed by the isocratic elution.

     
The main screen shows this accelerated separation with a perfectly flat baseline despite the steep gradient formation during the initial 30 minutes.   The inset screen shows a typical baseline using a competitor's detector that shows a distinct baseline incline during this relatively shallow gradient (60 mins) formation due to refractive index changes. The Cecil optical system of the CE4200 UV-Visible detector almost eliminates the sloping background.
